Evidence for McKusick’s hypothesis of deficient collagen in patients with homocystinuria

چکیده

Osteoporosis occurs commonly in homocystinuria. The underlying pathobiochemical mechanism remains unclear: disturbed cross-lin king of collagen has been suggested but this hypothesis has not been fully tested, nor have studies on collagen synthesis been performed. We therefore used recently available noninvasive tests for collagen synthesis and cross-linking to examine 10 patients with homocystin uria. Synthesis of collagen type I and type III was not different from age-matched healthy controls as reflected by comparable plasma levels of carboxyterminal propeptide of type I procollagen (PICP) and of plasma levels of N-terminal propeptide of procollagen type III (PIIINP). Collagen type I cross-links expressed by serum carboxyterminal telopeptide of collagen type I (ICTP) were 1.14 ± 0.24 ¿ig/1 in the patient group versus 3.29 ± 0.32 f i g / I in the control group. This significant reduction of cross-links in the group with homocystinuria did not correlate with serum homocysteine or homocysteie acid concentrations. Our data clearly indicate that the disturbed cross-linking hypothesis still holds and that the bone manifestations of homocystinuria are not due to deficient collagen synthesis.

Polymeric collagen of skin in normal sunjects and in patients with inherited connective tissue disorders.

1. The major collagen fraction of skin, which in vivo consists of tropocollagen molecules aggregated into extracellular fibres, has been extracted from forty-nine normal subjects and nineteen patients with inherited and acquired disorders of connective tissue. 2. In normal subjects the chemical stability of this fraction progressively increased up to the age of 60. 3. This fraction was less sta...

Hyperbilirubinemia Following Exchange Transfusion with G-6-PD Deficient Donor Blood

Background: The incidence of glucose-6-phosphate dehydrogenase (G-6-PD) deficiency in Iran is estimated at 10-14.9%.  The donor blood in blood banks is not screened routinely for this enzyme deficiency and such blood may be used for neonatal exchange transfusion. Objective: To study the effect of G-6-PD deficient blood in neonatal exchange blood transfusion. Methods: In a prospective study, serum...

Patients with homocystinuria have several connective tissue manifestations, the most severe being atherosclerosis. Furthermore, elevated plasma homocysteine concentrations correlate with atherosclerosis without other features of homocystinuria [1]. Homocysteine has been shown to interfere with collagen cross-linking, although the exact biochemical mechanism is unclear [2]. Moreover, the serum l...

A novel deletion and two recurrent substitutions on type VII collagen gene in seven Iranian patients with epidermolysis bullosa

Objective(s): Epidermolysis bullosa is one of the most important series of mechano-bullous heritable skin disorders which is categorized into four major types according to the layer that bullae forms within basement membrane zone. In dystrophic form of the disease, blisters are made in the sublamina densa zone, at the level of type VII collagen protein which produce anchoring fibrils. Type VII ...

Selective deficits in human audition: evidence from lesion studies

The human auditory cortex is the gateway to the most powerful and complex communication systems and yet relatively little is known about its functional organization as compared to the visual system. Several lines of evidence, predominantly from recent studies, indicate that sound recognition and sound localization are processed in two at least partially independent networks. Evidence from human...
